Assuming you are talking about FM transmitters? 

PA voltage x PA current x Eff (transmitter efficiency as listed in the transmitter manual). You'll have to assume that your line/antenna is 50 ohms.

Use the bird meter, and run the transmitter into a known 50 ohm dummy load ( since it will be a non-reactive, resistive load you can make sure the load is good by measuring it with a good ohmmeter.  Adjust the transmitter power until the Bird says you are at licensed TPO,  adjust the tx meter cal to make the tx read 100%.
